Electron Microscopic Study of Hepatotoxicity Induced by 2, 4-Dichlorophenoxyacetic Acid in Rats

ABSTRACT

Liver is a vital organ composed of many organelles such as mitochondria, endoplasmic reticulum, and lysosomes, each of which can be affected by toxicity. 2,4-dichlorophenoxyacetic acid (2,4-D), a phenoxy herbicide, has been studied to cause hepatotoxicity. This study was carried out using a total number of 25 male albino Wister rats grouped into control and 2,4-D LD50 treated group. Both groups were sacrificed after 24 hours. Liver tissue samples were obtained for histological study. Intoxicated animals in the 2,4-D group indicated marked alterations in the hepatocytes such as the existence of numerous lipid droplets, and the nucleus showed changes in nuclear shape and chromatin condensation, all of which are characteristic of an apoptotic cell. The nuclear membrane was inconspicuous. There was paucity of cytoplasmic organelles like rER, in addition to mitochondrial swelling and glycogen aggregates. The EM morphologic analysis of liver tissue demonstrated that 2,4-D has hepatotoxic potential and may cause hepatotoxicity.
