Antioxidant, Anti-inflammatory and Anti-proliferative potential of Amaranthus viridis and Swertia chirata

Abstract

Medical plants have been used to treat various chronic disease from ancient times. The development of science and technology in recent time makes it possible to discover the active constituents present in these plants and establish their biological targets for evolving them as a remedy for treating diseases.  The aim of this study was to determine the phytochemical constituents and antioxidant potential of methanolic extracts of Amaranthus viridis and Swertia chirata leaf as well as its ability to protect DNA from oxidative damage and anti-proliferative activity on B16F10 melanoma cell line. Phytochemical’s screening was done using standard qualitative method. The antioxidant activity was tested for both the methanolic extracts using DPPH assay and FRAP assay whereas anti-inflammatory potential was determined by RBC assay. Oxidative damage to DNA was induced using OH radical generated from UV/H2O2 radical system and checked on a 1.5 % agarose gel. In vitro anti-proliferative activity on B16F10 melanoma cell line was evaluated by MTT assay. Antioxidant capacities for both extracts were found to be good based on the tests performed. Both the extracts cause less than 50% lysis of RBC and thus it does not show considerable irritation potential.   A. viridis and S. chirata leaf extract exhibited a strong capability for DNA damage protection at 10 mg/ml concentration. The methanolic extract of A. viridis and S. chirata on B16F10 melanoma cell line showed that the percentage with regard to viability of cells was found to be 73.59% and 62.87% respectively at the concentration of 50μl/ml which decrease with the increase in concentration.
